Are you sure your dealing with lock nuts and not bolts? Which transmission are you playing with? The 4R44E and 4R55E both have studs that poke through the flex plate. If you didn't get the studs through the flex plate when installing the transmission, you have to pull the transmission back out and then make sure they are lined up with the holes in the flex plate as you slide the transmission up against the back of the engine. You also have to be sure the torque converter is fully engaged in the transmission's oil pump tangs. If not, you'll bind the torque converter when you tighten the bellhousing bolts or worse snap off part of the bellhousing. Since we volunteer our time and knowledge, we ask for you to please follow up when a problem is resolved.

Is that nut visible? If not tightened down and sticking past the studs it would snag on the housing when you turn it. If you can see it isn't touching anything and the torque convertor cannot turn at all by prying along the gear teeth it would be like DS says, the convertor wasn't splined in to the pump splines. It would have to be pulled back out to get set in properly. There are a few steps in getting the convertor splined on, and if you felt it go onto the first set and not seat all the way to the second you will have this issue. It can be frustrating and requires at times a patient and delicate touch to get them all the way in.
